Overview

Phosphodiesterase 4 (PDE4) isoforms are a family of enzymes that specifically hydrolyze cyclic adenosine monophosphate (cAMP), a key second messenger involved in regulating numerous physiological processes. PDE4 isoforms are widely expressed and play essential roles in inflammation regulation, neuroprotection, and metabolism. The PDE4 family consists of four genes: PDE4A, PDE4B, PDE4C, and PDE4D, each producing multiple isoforms with unique N-terminal regions that determine their specific subcellular localization. Different isoforms have distinct regulatory properties, including phosphorylation-dependent modulation and dimerization. PDE4 is a target for anti-inflammatory and immunomodulatory drugs, but current inhibitors lack subtype selectivity, leading to potential side effects.
