Phosphodiesterase 6 is an enzyme complex essential for the phototransduction cascade in vertebrate retinal photoreceptors[7]. It hydrolyzes cyclic GMP (cGMP), the second messenger responsible for keeping cGMP-gated ion channels open in darkness[1][7]. The PDE6 holoenzyme consists of two catalytic subunits (PDE6α and PDE6β in rods; combinations in cones) and two smaller inhibitory γ subunits (PDE6γ)[1][2][5][7]. Activation of PDE6 is mediated by the G protein transducin, which relieves inhibition by PDE6γ, allowing rapid hydrolysis of cGMP in response to light[1][5][7]. Structural studies reveal a complex architecture with regulatory GAF domains and finely tuned inter-subunit interactions that enable precise control of enzyme activity[1][4][5]. Mutations in PDE6 subunits are linked to various forms of inherited retinal degeneration, making it a critical molecular target in vision science and a potential target in retinal therapeutics[7]. Due to its structural similarity with PDE5, drugs like sildenafil and vardenafil developed for other targets can also inhibit PDE6, explaining certain drug-induced visual side effects[6][3].
