Overview

The phosphodiesterase enzyme family comprises key regulators of cell signaling via degradation of cyclic nucleotides. These enzymes catalyze the hydrolysis of phosphodiester bonds in cAMP and cGMP, thereby controlling the amplitude and duration of signaling pathways. The superfamily consists of 11 families (PDE1-PDE11), each with distinct substrate specificities, regulatory mechanisms, and tissue distributions. This diversity enables precise spatial-temporal control over numerous biological pathways—and makes them important drug targets across many therapeutic areas, including cardiovascular disease, inflammation, and neurological disorders.
