Phosphodiesterase III (PDE3) is an enzyme belonging to the phosphodiesterase family, which hydrolyzes cyclic nucleotides—specifically cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP)—into their inactive forms. PDE3 plays a critical role in regulating intracellular levels of these second messengers, thereby influencing various physiological processes. PDE3 consists of two main isoforms: PDE3A and PDE3B. PDE3A is primarily involved in cardiovascular function and fertility, while PDE3B is mainly implicated in lipolysis but also present in other tissues. Pharmacological inhibition provides therapeutic benefit acutely but carries significant risks chronically due to pro-arrhythmic potential.
