Overview

Phosphodiesterase isozyme 4 (PDE4) is a member of the phosphodiesterase enzyme family that hydrolyzes cAMP. It exists in four subfamilies: PDE4A, PDE4B, PDE4C, and PDE4D. By controlling intracellular cAMP levels, PDE4 regulates downstream signaling pathways influencing synthesis of pro- and anti-inflammatory cytokines, T cell activation, neutrophil degranulation, antigen presentation, and epithelial integrity. Inhibition of PDE4 leads to increased intracellular cAMP levels, activating PKA and modulating inflammatory responses. PDE4 is a therapeutic target for inflammatory disorders including pulmonary diseases and dermatological conditions.

Mechanism of action

Inhibition of PDE4 leads to increased intracellular cAMP levels, activation of PKA, and subsequent phosphorylation events affecting transcription factors such as CREB and NF-kappa B, ultimately modulating inflammatory responses.
