Overview

Phosphodiesterase type IV (PDE4) is a member of the phosphodiesterase enzyme superfamily, which catalyzes the hydrolysis of cyclic adenosine monophosphate (cAMP) to its inactive form. PDE4 is highly specific for cAMP and plays a crucial role in regulating intracellular levels of cAMP, thereby modulating various cellular responses to hormones, neurotransmitters, and other signaling molecules. It consists of four subtypes: PDE4A, PDE4B, PDE4C, and PDE4D. It is highly expressed in immune cells, epithelial cells, and brain tissue. Therapeutic targeting focuses on selective inhibition, but adverse gastrointestinal effects remain a challenge due to broad tissue distribution.
