Phosphoinositide 3-kinase catalytic subunit gamma isoform (PIK3CG), also known as PI3Kγ or p110-gamma, is a lipid kinase that plays a crucial role in cellular signaling, particularly within the immune system and cancer biology. It functions by phosphorylating phosphoinositides, thereby modulating extracellular signals and regulating cell proliferation and survival. PI3Kγ is particularly important for cytotoxicity regulation in natural killer (NK) cells and acts as a master regulator of immune cell function, especially within G protein-coupled receptor (GPCR) signaling pathways. It also participates in E-cadherin-mediated cell-cell adhesion and maintenance of epithelial integrity. Dysregulation or mutations of PIK3CG are implicated in various cancers, including intestinal, lung, and skin cancers, and it has been identified as a myeloid-restricted vulnerability factor in acute myeloid leukemia (AML). Targeting PIK3CG with inhibitors like IPI-549 has shown efficacy against AML progression.
