Phosphoinositide 3-kinase delta isoform (PI3Kδ) is a member of the class I PI3K family, primarily expressed in leukocytes and playing a central role in immune cell signaling. It is crucial for the development, differentiation, and function of various immune cells, including B cells and T cells. It catalyzes the phosphorylation of phosphatidylinositol-(4,5)-bisphosphate [PI(4,5)P2] to generate phosphatidylinositol-(3,4,5)-trisphosphate [PIP3], which acts as a membrane docking site for proteins with pleckstrin homology domains. Dysregulation contributes to diseases such as Activated PI3K-delta syndrome (APDS) and hematological malignancies, making it a target for therapeutic intervention.
