Overview

Phosphoinositide 3-kinases (PI3Ks) are a family of lipid kinases that phosphorylate the 3' hydroxyl group of phosphatidylinositol lipids. They play a crucial role in various cellular processes, including cell growth, proliferation, survival, motility, and intracellular trafficking. PI3Ks are classified into three main classes based on their structure and substrate specificity. The pathway is tightly regulated by opposing lipid phosphatases such as PTEN. Dysregulation of the PI3K pathway is implicated in cancer and other diseases, making it a therapeutic target.
