Phosphoinositide-3-kinase regulatory subunit 6 (PIK3R6) is a regulatory subunit of the phosphoinositide 3-kinase (PI3K) gamma complex, functioning primarily as an adapter protein that facilitates the activation of the PI3K gamma catalytic subunit (PIK3CG) by G protein beta-gamma dimers[1][2][4]. The PIK3CG:PIK3R6 heterodimer displays distinct regulatory properties compared to other PI3K gamma complexes, being less sensitive to activation by beta-gamma subunits and requiring HRAS association for optimal activity[1][2]. PIK3R6 mediates the recruitment of PI3K gamma to signaling complexes involved in processes such as angiogenesis and inflammatory signaling, and therefore plays important roles in many cellular processes, including cell signaling downstream of G protein-coupled receptors, cell proliferation, survival, differentiation, and immune response[1][2][3]. PIK3R6 and the broader PI3K pathway are implicated in disease processes such as cancer, inflammation, and cardiovascular dysfunction[3].
