Overview

Phosphoinositide kinase, FYVE-type zinc finger containing enzyme (PIKfyve) is a lipid and protein kinase that plays a crucial role in endomembrane homeostasis. It phosphorylates phosphatidylinositol 3-phosphate [PtdIns(3)P] to generate phosphatidylinositol (3,5)-bisphosphate [PtdIns(3,5)P₂], and also produces phosphatidylinositol 5-phosphate (PtdIns5P). These products regulate vesicular trafficking pathways and lysosome fission. Dysfunctions are linked to corneal dystrophy fleck type and potentially other cellular dysfunctions.

Mechanism of action

Inhibition of PIKfyve leads to impaired synthesis of PtdIns(3,5)P₂ and subsequent lysosome enlargement and cytoplasmic vacuolation.
