Overview

Phosphoinositide kinase FYVE-type zinc finger containing enzyme (PIKfyve) is a lipid kinase that phosphorylates phosphatidylinositol 3-phosphate (PtdIns(3)P) to generate phosphatidylinositol 3,5-bisphosphate [PI(3,5)P₂] and synthesizes phosphatidylinositol 5-phosphate [PI(5)P]. It plays a crucial role in regulating vesicular trafficking within the endosomal–lysosomal system, including lysosome fission and reformation, and retrograde transport. Disruption or inhibition leads to defects in membrane trafficking, ion homeostasis within lysosomes, vacuole/lysosome enlargement, impaired granule biogenesis, defective cell migration, and altered immune responses. Pharmacological inhibitors are used to probe roles in viral replication cycles and other cellular processes.
