Phospholipase A2 group IID (PLA2G2D) is a **secreted enzyme** part of the phospholipase A2 (sPLA2) superfamily that hydrolyzes the sn-2 acyl bond of phospholipids, yielding free fatty acids and lysophospholipids[3]. It is most highly expressed in lymphoid tissues and functions as a key regulator of immune responses by promoting anti-inflammatory lipid mediator synthesis, such as resolvins[4]. PLA2G2D dampens adaptive immune reactions and is involved in the resolution phase of inflammation, making it a potential target for therapeutic intervention in inflammatory and autoimmune diseases[4]. However, its precise physiologic and disease mechanisms are still being clarified. **Note:** Functions of group IID are better defined in animal models; in humans, research is ongoing and functions may partially overlap with other secreted PLA2 enzymes[3]. No widely approved drugs are clinically targeting PLA2G2D as of now; available inhibitors are primarily investigational tools.
