Phospholipase A2 group IVB (PLA2G4B) is a cytosolic enzyme belonging to the phospholipase A2 superfamily, primarily responsible for hydrolyzing the sn-2 acyl bond of phospholipids in cellular membranes and releasing free fatty acids and lysophospholipids. Unlike group IVA (cPLA2-alpha), PLA2G4B is less studied, with possible association with intracellular compartments such as mitochondria and early endosomes. Biological roles include modulation of lipid signaling pathways, which can influence inflammation and cell survival. Gene fusions (JMJD7–PLA2G4B) have been linked to oncogenic AKT pathway activation. Although considered a potential therapeutic target due to its enzymatic activity, drug development is predominantly focused on other PLA2 family members, and specific clinical applications for PLA2G4B remain undeveloped.
