Phospholipase A2 group IVD (PLA2G4D) is a member of the phospholipase A2 superfamily, specifically belonging to the cytosolic group IV enzymes. PLA2G4D is a calcium-dependent phospholipase A2 that selectively hydrolyzes the sn-2 position of membrane glycerophospholipids, with a preference for linoleic acid. This activity results in the release of free fatty acids and lysophospholipids, contributing to lipid signaling, membrane remodeling, and the production of bioactive lipid mediators. As a protein-coding gene, PLA2G4D is associated with pathways including glycerophospholipid biosynthesis and acyl chain remodelling. While general PLA2 enzymes are implicated in inflammatory processes and are considered drug targets, specific roles for PLA2G4D in disease have been documented for Oliver-McFarlane Syndrome and pityriasis rubra pilaris, but its broader therapeutic targeting remains underexplored.
No drugs specifically targeting PLA2G4D are documented. General mechanism for PLA2 family inhibitors involves inhibition of fatty acid release from membrane phospholipids, thereby reducing the synthesis of pro-inflammatory lipid mediators. Specific mechanisms for PLA2G4D-targeting drugs are not described in current literature.
