Overview

Phospholipase A2 receptor 1 (PLA2R1) is a transmembrane receptor that binds secretory phospholipase A2 enzymes, mediating diverse signaling events in immune cells and podocytes. Its principal clinical role is as the major antigenic target for autoantibodies in idiopathic membranous nephropathy. PLA2R1 participates in cell signaling pathways (MAPK cascade), endocytosis, regulation of cell survival, tissue homeostasis, and acts as a suppressor of tumorigenesis and cellular senescence. Genetic and serological biomarkers (anti-PLA2R1 antibodies, risk alleles) are used for disease diagnosis, prognosis, and monitoring; therapeutic strategies include B-cell targeted drugs and standard immunosuppression. The receptor is also implicated in inflammation, tissue injury responses, and may regulate podocyte adhesion in the kidney glomerular basement membrane.
