Phospholipase C beta 1 (PLCβ1) is an enzyme encoded by the PLCB1 gene in humans and is a critical component of cellular signal transduction pathways. It catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) into two key second messengers, inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG), thereby regulating intracellular calcium levels and activating protein kinase C signaling. PLCβ1 is activated by G protein-coupled receptors through Gαq and Gβγ subunits and is present in both cytoplasmic and nuclear compartments, reflecting diverse roles in cell proliferation, differentiation, and nervous system function. Its dysfunction is implicated in neurological disorders, certain cancers, and hematological diseases. Therapeutic modulation of PLCβ1, either directly or via associated pathways, remains challenging due to its centrality in key cellular processes and the risk of disrupting physiological homeostasis[1][2][4][5][6][7].
