Phospholipase C delta-4 (PLCD4) is an enzyme in the delta subclass of phosphoinositide-specific phospholipase C family. It plays an essential role in hydrolyzing phosphatidylinositol 4,5-bisphosphate (PIP2) to generate second messengers, diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3)[1][4]. These messengers activate protein kinase C and release calcium from intracellular stores, respectively, making PLCD4 a key regulator of intracellular signaling, especially calcium-mediated events. PLCD4 is especially important for the acrosome reaction in sperm during fertilization and plays significant roles in cell proliferation, the cell cycle (notably G1/S transition), and may contribute to tumorigenesis when dysregulated. Predominant expression is noted in brain, testes, and regenerating liver, with functional evidence for contributions to both cytoplasmic and nuclear calcium signaling, and possible roles in cellular senescence and oncogenesis[2][5][7].
