Phospholipase C eta-2 (PLCH2) is a neuron-specific member of the phosphoinositide-specific phospholipase C (PLC) superfamily of enzymes, which catalyze the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) into the second messengers inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG), mediating key steps in intracellular signal transduction[1][2][3][5]. PLCH2 is characterized by structural domains, including a pleckstrin homology (PH) domain, EF-hand motifs, catalytic X and Y domains, and a unique C-terminal region[1][2]. PLCH2 activity is highly sensitive to calcium and may be stimulated by Gβγ subunits of G-protein-coupled receptors[1]. Expression of PLCH2 is mostly restricted to neurons (particularly in the hippocampus, cortex, and olfactory bulb) and is implicated in the formation and maintenance of neuronal networks during brain development[1][2]. Functional studies have shown that PLCH2 is essential for neurite outgrowth and interacts with LIMK-1, a key cytoskeletal regulator, highlighting its role in neuronal differentiation and actin cytoskeleton organization[2]. There are currently no known drugs or biomarkers directly targeting PLCH2, nor clear safety concerns described in the literature.
