Phospholipid phosphatases (PLPPs) are a family of integral membrane enzymes that catalyze the dephosphorylation of various lipid phosphate substrates, including lysophosphatidic acid (LPA), sphingosine-1-phosphate (S1P), and phosphatidic acid. These enzymes play crucial roles in regulating lipid signaling pathways, cellular uptake of bioactive lipids, and maintaining lipid homeostasis. Alterations in PLPP expression or function can impact numerous physiological processes, and dysregulation is linked with cancer progression due to effects on cell proliferation/migration pathways. In infectious disease contexts, some pathogens secrete proteins with phospholipid phosphatase activity that modulate host immune responses.
