Overview

Phospholipid phosphatase 1 (PLPP1) is a magnesium-independent integral membrane enzyme of the PAP2 (type II phosphatidic acid phosphatase) family[1]. It catalyzes the dephosphorylation of a variety of lipid phosphate substrates, including phosphatidate, lysophosphatidate, sphingosine 1-phosphate, and ceramide 1-phosphate. PLPP1 regulates the balance of these bioactive lipids in the extracellular space and at the plasma membrane, thereby influencing signal transduction in processes such as cell proliferation, migration, inflammation, and platelet activation. Its dysregulation has been implicated in pathologies such as cancer and inflammatory diseases. Expression levels of PLPP1 can serve as a prognostic marker in breast cancer and may affect tumor progression or relapse[1][3]. PLPP1 is not currently a routine clinical drug target, but modulation of its enzymatic activity is of increasing research interest in oncology and metabolic disorders[3].
