Phospholipid phosphatase 2 (PLPP2) is a membrane-associated enzyme in the phospholipid phosphatase family that catalyzes the dephosphorylation of a broad range of bioactive lipid phosphates including lysophosphatidate (LPA), phosphatidate (PA), and sphingosine 1-phosphate (S1P)[1]. PLPP2 is encoded by the PPAP2C gene and structurally characterized by multiple transmembrane domains. It plays an essential role in the regulation of cell signaling through the metabolism of lipid substrates and is widely expressed across human tissues. In cancer, especially breast cancer, PLPP2 is frequently overexpressed and promotes tumor growth and progression by enhancing cell proliferation, migration, and invasion while associating with the cell-cycle regulator c-Myc[2][4][6]. Knockout or knockdown studies demonstrate that reducing PLPP2 levels inhibits tumor growth, implicating PLPP2 as a potential therapeutic target, though no drugs currently target it directly[4][6]. Elevated PLPP2 expression is a negative prognostic indicator in several cancers, particularly in ER+ breast cancer[2][4].
