Phospholysine phosphohistidine inorganic pyrophosphate phosphatase (LHPP) is a hydrolase enzyme that catalyzes the removal of phosphate groups from phospholysine, phosphohistidine, and inorganic pyrophosphate, acting on both nitrogen-phosphorus and oxygen-phosphorus bonds. LHPP is considered a member of the haloacid dehalogenase (HAD)-like hydrolase family and functions as a homodimer, localized primarily in the cytosol and nuclear specks. It plays a crucial role in maintaining cellular phosphate homeostasis and acts as a tumor suppressor, with reduced expression observed in various cancers. The tumor-suppressive effects of LHPP involve inhibition of key oncogenic signaling pathways, regulation of cell cycle, and promotion of apoptosis. It does not currently have known interacting drugs or approved small-molecule modulators, but LHPP and its pathways are under study for their prognostic and therapeutic relevance, particularly as a biomarker and therapeutic target in oncology.
