Overview

Phosphoribosyl pyrophosphate amidotransferase (PPAT) is a key regulatory enzyme that catalyzes the first committed step in de novo purine nucleotide biosynthesis. It converts 5-phosphoribosyl-1-pyrophosphate (PRPP) and glutamine into 5-phosphoribosylamine (PRA), glutamate, and pyrophosphate. Elevated expression or activity is associated with increased cell proliferation and cancer progression, particularly in thyroid cancer. It regulates PKM2 expression and activates ERK/STAT3 pathways involved in tumorigenesis.
