Overview

Phosphoribosylformylglycinamidine synthase (PFAS) is an enzyme that catalyzes a key ATP-dependent step in the de novo biosynthesis of purine nucleotides. It converts N2-formyl-N1-(5-phospho-D-ribosyl)glycinamide (FGAR), L-glutamine, and water into 2-(formamido)-N1-(5-phospho-D-ribosyl)acetamidine (FGAM), L-glutamate, ADP, and phosphate. PFAS is essential for purine metabolism, which underpins DNA replication, transcription, energy metabolism, and other cellular processes requiring nucleotides. PFAS activity can be regulated by phosphorylation events mediated by ERK signaling pathways and via PW1 expression during cardiac injury.
