Phosphoserine phosphatase (PSPH) is a magnesium-dependent enzyme that catalyzes the final and irreversible step in the de novo biosynthesis of L-serine. It hydrolyzes O-phospho-L-serine to produce L-serine and inorganic phosphate. L-serine serves as a precursor for proteins, nucleotides, sphingolipids, and neuromodulators like D-serine and glycine, which are essential for brain development and function. Mutations in PSPH can cause Neu-Laxova syndrome and phosphoserine phosphatase deficiency, characterized by severe neurological symptoms due to impaired serinogenesis.
