Overview

Photon energy is a fundamental physical property representing the energy carried by a single photon, determined by the equation E=hf, where h is Planck's constant and f is the frequency [1]. In biological contexts, it is not a therapeutic target such as a protein or enzyme, but rather a physical stimulus that initiates biochemical cascades by interacting with chromophores and photoreceptors like rhodopsin or melanopsin [2]. Clinically, specific levels of photon energy (wavelengths) are utilized in photodynamic therapy (PDT) to activate photosensitizing drugs, facilitating the targeted destruction of malignant cells through the generation of reactive oxygen species [3]. Additionally, the absorption of solar photon energy in the ultraviolet B (UVB) range is essential for the cutaneous synthesis of Vitamin D, although excessive energy in the UV spectrum can lead to direct DNA damage and the development of skin neoplasms [4,5]. Because photon energy is a measurement of electromagnetic radiation rather than a biological molecule, it is classified as 'incorrect' when referred to as a pharmacological target.

Mechanism of action

Photon energy is not a target itself but acts as an activating stimulus for photosensitizing drugs or endogenous receptors. In photodynamic therapy, photons of specific wavelengths excite photosensitizers to a triplet state, which kemudian transfers energy to molecular oxygen, generating cytotoxic singlet oxygen and reactive oxygen species [3].
