Overview

A physical barrier to soft tissue ingrowth is a mechanical concept used in Guided Tissue Regeneration (GTR) and Guided Bone Regeneration (GBR) rather than a specific molecular receptor or protein target. This strategy involves placing a biocompatible membrane over a bone or periodontal defect to physically prevent the migration of fast-growing soft tissue cells, such as fibroblasts and epithelial cells, into the wound space [PMID: 28932971]. Because bone cells migrate and proliferate significantly slower than soft tissue cells, the absence of a barrier often leads to the defect being filled with fibrous connective tissue rather than functional bone [PMID: 30141384]. In clinical practice, these barriers are achieved using materials like collagen (often Type I or III), which may be resorbable, or non-resorbable materials like expanded polytetrafluoroethylene (ePTFE) and titanium [PMID: 23640009]. While these barriers do not interact with traditional small-molecule drugs through receptor-ligand binding, they serve as a critical "target" for physical intervention in reconstructive dentistry and orthopedics. Modern advancements also explore "bioactive" barriers that can deliver growth factors like Bone Morphogenetic Proteins (BMPs) to further enhance the regenerative process [PMID: 29243762].
