Overview

The physical microenvironment, or tissue compartments, encompasses the structural and mechanical aspects of tissues that surround and interact with cells. This includes the extracellular matrix (ECM), various cell types (such as fibroblasts, immune cells, endothelial cells), and the physical properties of these components—such as stiffness, porosity, fiber alignment, and topography. These features collectively define distinct tissue compartments within organs or tumors. It provides structural support, establishes physical boundaries, regulates signaling, and modulates diffusion. The tumor microenvironment’s physical properties influence tumor progression by affecting proliferation, stemness, migration, invasion, and metastasis. Cells sense their physical environment through mechanotransduction pathways. Targeting the stroma in cancer therapy aims to disrupt these supportive interactions.
