Physiologic ion homeostasis refers to the complex regulatory processes that maintain stable concentrations of essential ions, such as sodium, potassium, calcium, and chloride, within intracellular and extracellular compartments (StatPearls, 2023). This balance is critical for fundamental cellular activities, including membrane potential maintenance, signal transduction, muscle contraction, and osmotic regulation (NCBI, 2022). Disruption of this homeostasis is linked to numerous pathological conditions, including cardiovascular diseases, neurological disorders, and renal failure (NIH, 2023). While not a single molecular target itself, it is the physiological outcome managed by various classes of drugs that target specific ion channels, transporters, and ATP-driven pumps (PubMed, 2021). For example, diuretics modulate sodium and water balance in the kidneys, while calcium channel blockers regulate vascular tone by influencing calcium entry into smooth muscle cells (StatPearls, 2023). Understanding the systemic regulation of these ions is vital for developing therapies that correct imbalances without causing off-target toxicity.
