Overview

Physiological and Metabolic Pathways refers to the vast network of biochemical reactions and systemic processes that sustain life, including energy production, nutrient processing, and signal transduction (National Center for Biotechnology Information, 2023). These pathways are composed of numerous individual enzymes, receptors, and transporters that maintain homeostasis and allow organisms to respond to environmental changes. While specific components within these pathways—such as HMG-CoA reductase in cholesterol synthesis—are well-validated therapeutic targets, the term "Physiological and Metabolic Pathways" itself is a broad biological category rather than a discrete molecular target (StatPearls, 2023). In drug discovery, targeting a specific node within a pathway is necessary to achieve therapeutic efficacy while minimizing off-target effects. Because this term encompasses nearly all biological activity, it is considered too broad to be classified as a single target for drug development. Disruption or dysregulation of these pathways is a hallmark of many diseases, including metabolic syndrome, diabetes, and various forms of cancer (Reactome, 2024). Consequently, while the study of these pathways is fundamental to pharmacology, the term does not represent a specific site of drug action.
