Overview

The physiological coagulation system is a highly regulated biological pathway essential for maintaining vascular integrity and preventing excessive blood loss following injury (StatPearls, 2023). It operates through a complex cascade of zymogen activations, primarily involving serine proteases known as clotting factors, which culminate in the generation of thrombin and the conversion of soluble fibrinogen into an insoluble fibrin mesh (NIH, 2022). This system is traditionally divided into the intrinsic, extrinsic, and common pathways, though modern models emphasize the cell-based nature of these interactions on platelet surfaces (Wikipedia, 2024). Beyond its primary role in hemostasis, the coagulation system is intricately linked to inflammatory responses and wound healing processes. Dysregulation of this system is a central feature in numerous life-threatening pathologies, including venous thromboembolism, myocardial infarction, and various bleeding disorders like hemophilia (PubMed, 2021). Consequently, many widely prescribed cardiovascular drugs, such as direct oral anticoagulants (DOACs) and heparins, function by targeting specific enzymatic components within this system to modulate its activity and prevent pathological clot formation.
