Physiological ion homeostasis is the highly regulated process of maintaining stable concentrations of essential ions, such as sodium, potassium, calcium, and chloride, within intracellular and extracellular compartments (StatPearls, 2023). This equilibrium is vital for maintaining membrane potential, driving signal transduction, and ensuring proper muscle and nerve function (NCBI Bookshelf, 2022). While 'ion homeostasis' is a physiological state or process rather than a single molecular target, it is governed by a diverse set of proteins including ion channels, transporters, and ATP-dependent pumps (Nature Reviews Molecular Cell Biology, 2019). Many therapeutic agents, such as diuretics, anti-arrhythmics, and calcium channel blockers, exert their effects by targeting these specific proteins to treat conditions like hypertension, heart failure, and neurological disorders (PubMed, 2021). Failure to maintain this balance can result in severe clinical manifestations, including cardiac arrhythmias, seizures, and metabolic disturbances (NIH, 2023).
Drugs modulate physiological ion homeostasis by targeting specific molecular components such as voltage-gated or ligand-gated ion channels, ATP-dependent ion pumps (e.g., Na+/K+-ATPase), and solute carriers/transporters to alter the flux of ions across biological membranes (StatPearls, 2023).
