Overview

Physiological state refers to the collective functional status of an organism's biological systems at a specific point in time, governed by homeostatic mechanisms and environmental interactions. It is not a discrete molecular target, such as a receptor, enzyme, or ion channel, but rather a descriptive term for the overall condition of a biological system (IUPHAR/BPS Guide to Pharmacology). In pharmacology, therapeutic agents are designed to interact with specific molecular targets to shift a pathological physiological state toward a healthy one. Because 'Physiological State' encompasses a vast array of interconnected pathways and systemic variables, it cannot be targeted with the molecular precision required for drug development. It serves as a phenotypic readout of drug action rather than the primary site of drug binding (Nature Reviews Drug Discovery, 2006). Examples of physiological states include sleep, wakefulness, fasting, or various disease states like hypertension. Monitoring these states is crucial for assessing drug efficacy and safety during clinical trials. However, the term itself lacks the specificity needed to be classified as a drug target in a database.
