Overview

Piezo-type mechanosensitive ion channel component 2 (PIEZO2) is a large transmembrane ion channel that forms a homotrimeric structure and serves as a primary mechanotransducer in sensory neurons, particularly Merkel cells and proprioceptors[4][7][9]. It is responsible for converting mechanical stimuli—such as light touch, vibration, and stretch—into electrical signals, thereby enabling the senses of touch and proprioception[5][6][9]. PIEZO2 conducts a nonselective cation current, with mild selectivity for Ca^2+, and is characterized by a high number of transmembrane regions and a unique propeller-like architecture[4][7]. Pathogenic mutations in the PIEZO2 gene are linked to several inherited disorders affecting movement and joint development, including distal arthrogryposis and Marden-Walker syndrome[4]. Disruption of PIEZO2 in animal models leads to profound impairments in coordination and touch sensation. Currently, there are no specific therapeutic drugs approved for targeting PIEZO2, but selective blockers such as GsMTx4 inhibit both PIEZO1 and PIEZO2[1]. Safety concerns when targeting PIEZO2 include interference with vital sensory functions such as touch and proprioception.
