PIEZO1 antisense RNA 1 (PIEZO1-AS1) is a long non-coding RNA transcribed in the antisense direction at the PIEZO1 gene locus. By its antisense nature, PIEZO1-AS1 may regulate expression of the PIEZO1 mechanosensitive ion channel, potentially via mechanisms such as transcriptional interference, formation of RNA-RNA hybrids, or epigenetic modulation of chromatin structure near the PIEZO1 gene. Unlike PIEZO1, which encodes a clinically significant mechanosensitive ion channel involved in vascular biology, cellular mechanotransduction, and hereditary diseases, PIEZO1-AS1 itself has no known enzymatic, receptor, transporter, or direct signaling activity, and does not serve as a therapeutic drug target at this time. The broader class of antisense RNAs is essential for fine-tuning gene expression in eukaryotes, but individual disease or drug relevance for PIEZO1-AS1 has not been established.
