PiggyBac transposable element derived 2 (PGBD2) is a member of the piggyBac-derived transposase gene family, which originated from mobile genetic elements. In humans, it encodes a transposase-like protein derived from the piggyBac transposon of the moth *Trichoplusia ni*. PGBD2, like others in the family (PGBD1–5), is believed to be a domesticated transposase but shows no obvious relationship to other known protein families, and its exact biological function remains unknown[1][2][4][5]. It is highly expressed in syncytiotrophoblasts, specialized placental cells[4], but there is limited evidence on its functional role, disease associations, or pharmacological relevance. There are currently no known drug interactions, mechanisms of action, or biomarker roles for PGBD2. The protein appears to be novel, with no conservation of the catalytic DDE/D domain characteristic of active transposases, indicating a loss of transposase activity and suggesting that its domestication was not coupled with the retention of this catalytic function[2]. Two transcript variants encoding different isoforms have been identified. Despite broad expression, no clear physiological or pathogenic function has been documented[1][4].
