Overview

PiggyBac transposable element-derived protein 4 (PGBD4) is part of the piggyBac transposase family, which originates from a transposon found in diverse animal genomes including humans. It is a human protein of approximately 67 kDa, and while its ancestral transposase function is evident from sequence homology, the PGBD4 protein itself does not act as a canonical transposase, nor does it show obvious transpositional or enzymatic activity in humans. Its molecular and cellular functions remain largely uncharacterized, with no clear biological pathways or disease mechanisms established as of current knowledge. Some data link variation in PGBD4 to neurodevelopmental disorders with eye movement abnormalities, but its mechanistic role, if any, in disease is undetermined. There is no evidence for its use as a therapeutic target, nor is there structured clinical knowledge about its safety, biomarkers, or small molecule interactions.
