Overview

PIK3CD antisense RNA 2 (PIK3CD-AS2) is a cytoplasmic long non-coding RNA transcribed antisense to the PIK3CD gene. It regulates cellular processes by modulating the stability of YBX1, a negative regulator of p53, thereby suppressing p53 signaling and promoting cell proliferation while inhibiting apoptosis. PIK3CD-AS2 is upregulated in lung adenocarcinoma, especially in p53 wild-type contexts, and is associated with poor patient prognosis. Preclinical inhibition of PIK3CD-AS2 using locked nucleic acid oligonucleotides exhibits tumor suppressive effects. Beyond cancer, PIK3CD-AS2 is differentially expressed in human brain tissue and patient-derived cells from schizophrenia, particularly in fetal brain, implicating it in neurodevelopmental regulatory networks. Additionally, it has been shown to interact with viral proteins such as H5N1-NS1, highlighting potential relevance in infection biology
