Overview

PilY1 is a multifunctional protein in *Pseudomonas aeruginosa* essential for type IV pili (T4P) assembly and function. It regulates twitching motility, surface adhesion, and host cell interaction. Acting as an adhesin at the pilus tip, PilY1 also functions as a mechanosensor, detecting mechanical forces. It modulates intracellular signaling pathways such as c-di-GMP production, controlling biofilm formation and other surface behaviors. PilY1 contains a vWA-like domain, a β-propeller domain, and an RGD motif for integrin binding, making it a key virulence factor.
