PINK1 antisense RNA 1 (PINK1-AS) is a human long non-coding RNA (lncRNA) transcribed from the region antisense to the PINK1 protein-coding gene. It is also known as FLJ00387, PINK1AS, naPINK1, NAPINK1, and PINK1-AS1. PINK1-AS is expressed in tissues rich in mitochondria such as skeletal muscle and exists as a natural antisense transcript (NAT) that overlaps with the PINK1 gene locus. Experimental data show that PINK1-AS modulates the expression of a PINK1 splice variant (svPINK1), likely by stabilizing or promoting its abundance, and thus indirectly regulates mitochondrial function. This regulatory role suggests that PINK1-AS may influence susceptibility or progression of diseases linked to PINK1 dysfunction, including early-onset Parkinson’s disease. There are currently no therapeutic agents or approved drugs that directly target PINK1-AS, and it is not an established therapeutic target. However, its biological effect on the expression of a critical mitochondrial kinase locus elevates the importance of PINK1-AS for understanding lncRNA-mediated regulation of disease-relevant genes[1][2][3].
