Overview

Crotalinae snake venom toxins are a complex mixture of bioactive proteins and peptides produced by pit vipers, including rattlesnakes (Crotalus), lanceheads (Bothrops), and copperheads (Agkistrodon). These toxins evolved to immobilize prey and initiate digestion by disrupting the circulatory, nervous, and muscular systems of the victim. Major components include snake venom metalloproteinases (SVMPs) that degrade the extracellular matrix leading to hemorrhage, serine proteinases (SVSPs) that interfere with the coagulation cascade, and phospholipases A2 (PLA2) that cause extensive tissue necrosis and myotoxicity (UniProt, 2024; PubMed, 2021). In clinical medicine, these toxins are the primary targets for antivenom therapies, which utilize purified antibody fragments to neutralize systemic toxicity. Beyond their role in envenomation, specific Crotalinae toxins have served as structural templates for the development of major pharmaceutical classes, such as ACE inhibitors derived from bradykinin-potentiating peptides and antiplatelet drugs like eptifibatide derived from disintegrins (NIH, 2022).

Mechanism of action

Antivenoms function through passive immunization, where venom-specific antibody fragments (Fab or F(ab')2) bind to and neutralize the enzymatic and toxic activities of venom components, preventing their interaction with host tissues and facilitating their systemic clearance (StatPearls, 2023; FDA, 2018). Small molecule inhibitors like Varespladib specifically target the active site of Phospholipase A2 to inhibit its catalytic and non-catalytic toxic effects (Lewin et al., 2016).
