Overview

Pituitary tumor-transforming gene 1 protein (PTTG1, also known as securin) is a natively unfolded protein that serves as a central regulator of sister chromatid separation during mitosis[1][2][3][4]. It prevents premature separation by inhibiting separase, a protease responsible for cleaving the cohesin complex, and is degraded by the anaphase-promoting complex (APC) at the metaphase–anaphase transition, enabling chromosome segregation[1][3][4]. PTTG1 also has key roles in transcriptional regulation, cell-cycle progression, tumorigenesis, and DNA repair, interacting with proteins like p53, Ku70, and DNAJA1[1][2]. Overexpression is linked to oncogenic transformation and is frequently found in tumors, but depending on context (e.g., mammary gland development), it may also function as a tumor suppressor[4]. Clinical interest centers on its role both as a biomarker and a potential cancer therapeutic target, though selective pharmacologic inhibition has yet to be achieved[1][2][3][4].
