Overview

Pituitary tumor-transforming gene 1 protein-interacting protein (PTTG1IP or PBF) is a single-pass type I integral membrane protein encoded on chromosome 21q22.3 that directly binds the proto-oncogene PTTG1, promoting its nuclear import and driving transcriptional activation of downstream targets such as basic fibroblast growth factor. PTTG1IP is expressed in many tissues but is notably upregulated in pituitary tumors, various grades of breast cancer, and thyroid lesions, associated with increased cell proliferation, invasion, and tumor recurrence. It also regulates the sodium-iodide symporter in thyroid cells, making it relevant in radioiodine uptake and thyroid cancer therapy. Although its physiological functions remain incompletely characterized, its links to tumor biology have highlighted it as a putative therapeutic or prognostic target in cancer research[1][3][5][7].
