Overview

Pituitary tumor-transforming gene 2 protein (PTTG2) is a member of the securin protein family and a close homolog of PTTG1 (Securin)[1][2][5]. PTTG2 is encoded by an intronless gene and displays high amino acid sequence similarity to PTTG1, but unlike PTTG1, it does not bind separase and thus is not involved in sister chromatid separation during mitosis[1][2]. PTTG2 functions are not fully characterized; current evidence indicates it plays a role in cell adhesion, regulation of epithelial-mesenchymal transition (EMT), apoptosis (cell death), and possibly cell invasion in cancer contexts[1][2][3]. Its expression is generally very low in most normal tissues but can be upregulated in tumors, where it may contribute to tumor progression and invasiveness[3][4][5]. While mechanisms have not been precisely delineated, PTTG2 is implicated in cancer biology, yet is not an established therapeutic target and no drugs specifically target PTTG2[2][3][4][5].
