Overview

PITX2 adjacent non-coding RNA (PANCR) is a long intergenic non-coding RNA (lncRNA) discovered adjacent to the PITX2 gene at the chromosome 4q25 locus[1][4]. PANCR is specifically expressed in the left atrium and eye, and not in other heart chambers[1][4]. It is co-expressed with PITX2c, a key isoform of the PITX2 transcription factor involved in cardiac development, and its levels are tightly correlated with those of PITX2c[1][4]. PANCR knockdown reduces PITX2c expression in differentiating cardiomyocytes, influencing the transcriptome similarly to direct knockdown of PITX2c, suggesting PANCR acts as a regulatory RNA controlling PITX2c expression, possibly by stabilizing its mRNA in the cytoplasm[1][4]. Observational studies associate variation at the PANCR/PITX2 locus with risk for atrial fibrillation, but PANCR itself is not currently a direct therapeutic target nor does it have known specific drug interactions or safety concerns[1][4].
