PKMYT1 associated long non-coding RNA (PKMYT1AR) is a human-specific lncRNA (also known as ENST00000595422) that is overexpressed in non-small cell lung cancer (NSCLC) and is associated with poor clinical outcomes. It is transcriptionally activated by the Yin Yang 1 (YY1) transcription factor and predominantly localized in the cytoplasm. PKMYT1AR acts as a ceRNA by sponging miR-485-5p, leading to upregulation of the protein kinase PKMYT1, which in turn enhances Wnt/β-catenin signaling and cancer stem cell maintenance by inhibiting the ubiquitin-mediated degradation of β-catenin. Knockdown or inhibition of PKMYT1AR impairs tumor cell proliferation, migration, xenograft tumor formation, and cancer stem cell phenotypes in cell and animal NSCLC models. This molecule is not a protein but a regulatory lncRNA, tightly coupled to cancer biology as a potential diagnostic, prognostic, and therapeutic target in lung cancer and potentially in other cancers where the PKMYT1/β-catenin/Wnt axis is relevant.
Acts as a competitive endogenous RNA (ceRNA) for miR-485-5p, upregulating PKMYT1 expression by sequestering miR-485-5p and thereby relieving its repression on PKMYT1. Promotes Wnt/β-catenin pathway signaling by stabilizing β-catenin through downstream effects on PKMYT1.
