Overview

PKNOX2 antisense RNA 1 (PKNOX2-AS1) is a long non-coding RNA situated opposite to the coding sequence of PKNOX2. Studies show that PKNOX2-AS1 can suppress the secretion of pro-inflammatory cytokines (like IL-1β, IL-6, IL-18, and TNF-α) and protect renal epithelial cells from damage and cell death induced by sepsis or inflammation. It achieves this by inhibiting NF-κB pathway activation and modulating chromatin structure, thus regulating gene expression at both transcriptional and post-transcriptional levels. This RNA molecule has no protein product and acts mainly through RNA-mediated mechanisms including miRNA sequestration and transcriptional modulation. Its downregulation is associated with increased susceptibility to inflammation-induced injury. Although research is at a preliminary stage, PKNOX2-AS1 is being considered for biomarker development and potential therapeutic intervention in conditions such as acute kidney injury and possibly cancer.
