Overview

Placenta-expressed transcript 1 protein (PLET1) is a cell surface protein encoded by the PLET1 gene (also known as C11orf34) that is involved in processes such as cell migration, wound healing, cell-matrix adhesion, and trophoblast differentiation[3][7][1]. It exhibits a highly regulated expression pattern, being tightly repressed by DNA methylation in embryonic stem cells and expressed in trophoblast stem cells and certain differentiated cells[1][3]. PLET1 plays a critical role in balancing trophoblast self-renewal and differentiation, influencing lineage specification towards trophoblast giant cells or syncytiotrophoblast cells[1]. It is predicted to localize in the extracellular region and plasma membrane, and acts as a marker for progenitor/stem cell reservoirs[1][2][3][7]. Diseases associated with dysregulation of PLET1 include arthrogryposis multiplex congenita and DiGeorge syndrome; however, there are currently no known drugs targeting this protein[3][6].
