Placental Growth Factor (PlGF)

Target
PlGF
Molecular classification
Growth Factor, VEGF sub-family, Secreted Protein
01

Overview

Placental growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family, crucial for angiogenesis and vasculogenesis, especially during embryonic development and pregnancy. It is primarily produced by placental trophoblasts. PlGF regulates blood vessel formation within the placenta, supports trophoblast growth and differentiation, and facilitates remodeling of maternal spiral arteries. Low levels are associated with preeclampsia and placental dysfunction. While not essential under normal conditions, it becomes critical during pathological states like ischemia or tumorigenesis.

Mechanism of action

PlGF binds primarily to VEGFR-1 (Flt-1), influencing biological responses. sFlt-1 antagonizes PlGF activity.

Safety considerations

Therapeutic targeting of PlGF requires careful consideration due to its role in normal angiogenesis and wound healing. Complete inhibition may lead to adverse effects.Altered angiogenic response under stress conditions needs to be considered when manipulating PlGF pathways.
